In clinical labs, sodium nitroprusside is a key reagent in nitroprusside tests for urinary ketone bodies and aminoaciduria, forming characteristic purple/red complexes with sulfhydryl‑containing compounds.

It is also used in some colorimetric methods as a catalyst or indicator, for example in phenate‑based ammonia determinations in water analysis.
